GFB-204 is a small molecule inhibitor of the Transient Receptor Potential Canonical 5 (TRPC5) ion channel, originally developed by Goldfinch Bio. It was designed as a targeted therapy for proteinuric kidney diseases, including focal segmental glomerulosclerosis (FSGS) and diabetic nephropathy. The drug's mechanism of action involves blocking the overactivation of TRPC5 channels in podocytes, which is a key driver of cytoskeletal disruption, foot process effacement, and subsequent protein loss in the urine. GFB-204 was positioned as a next-generation or backup compound to Goldfinch's lead candidate, GFB-887. Following the dissolution of Goldfinch Bio in early 2023 and the acquisition of its TRPC5 program by Karuna Therapeutics (now part of Bristol Myers Squibb), GFB-204 is no longer being actively developed, as the focus has shifted to the lead asset.
