Overview

GFB-8438 is a potent and subtype-selective small molecule inhibitor of the transient receptor potential canonical channel 5 (TRPC5), with additional activity against TRPC4. Developed by Goldfinch Bio, the compound was specifically designed for the treatment of proteinuric kidney diseases, such as focal segmental glomerulosclerosis (FSGS). By inhibiting TRPC5, GFB-8438 aims to protect podocytes from injury and reduce the leakage of proteins into the urine. Preclinical studies in mouse podocytes and rat models of hypertensive kidney disease demonstrated significant reductions in proteinuria and albuminuria. However, following the cessation of operations at Goldfinch Bio, the development of GFB-8438 has been discontinued.
