Overview

GFB-887 is a podocyte-targeting, small molecule inhibitor of transient receptor potential canonical-5 (TRPC5), developed as a precision medicine for the treatment of glomerular kidney diseases characterized by overactivation of the TRPC5-Rac1 pathway. By inhibiting TRPC5, GFB-887 aims to protect podocytes and prevent proteinuria, which is a hallmark of progressive kidney diseases such as focal segmental glomerulosclerosis (FSGS), treatment-resistant minimal change disease (TR-MCD), and diabetic nephropathy (DN). Clinical studies have shown that GFB-887 reduces urinary Rac1 concentrations in a dose-dependent manner, confirming target engagement and suppression of the pathogenic pathway. The drug has demonstrated safety and tolerability in Phase 1 trials and showed clinically meaningful reductions in proteinuria in Phase 2 studies for FSGS. It is administered orally once daily[1][4][6][7].
