Overview

GFH312 is a small molecule, oral inhibitor of receptor-interacting serine/threonine-protein kinase 1 (RIPK1), a key regulator in the TNF-alpha signaling pathway. By inhibiting RIPK1, GFH312 blocks necroptosis—a form of programmed cell death associated with inflammation—and reduces inflammatory responses. The drug has demonstrated central nervous system penetration and target engagement in early clinical studies, as evidenced by decreased phospho-RIPK1 levels in peripheral blood mononuclear cells and cerebrospinal fluid exposure above the inhibitory concentration for necroptosis. GFH312 is being developed primarily for autoimmune diseases and peripheral vascular diseases, including peripheral artery disease (PAD) and intermittent claudication. It is also under investigation for cholangitis and rheumatoid arthritis. The developer is GenFleet Therapeutics[2][3][4][5][6][7].
