GFPT1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the GFPT1 gene, which encodes glucosamine-fructose-6-phosphate aminotransferase, the rate-limiting enzyme of the hexosamine biosynthetic pathway. In preclinical research, particularly in nasopharyngeal carcinoma (NPC) and head and neck cancer models, GFPT1 silencing has been shown to reduce the production of TGF-β1. This reduction in TGF-β1 levels enhances the sensitivity of cancer cells to ionizing radiation by increasing radiation-induced DNA double-strand breaks and reducing the clonogenic capacity of the tumor cells. The University of Hong Kong has explored this siRNA as a potential radiosensitizer to improve the prognosis of NPC patients and reduce the toxicity associated with high-dose radiotherapy.
