GH55 + GH21 is an investigational combination therapy developed by Suzhou Genhouse Bio Co., Ltd. for the treatment of advanced solid tumors harboring aberrantly activated MAPK signaling. GH55 is a small molecule, dual-mechanism inhibitor of ERK1/2 that targets both the kinase activity and its activation by MEK, potentially overcoming resistance mechanisms. GH21 (also known as HBI-2376) is a potent, orally active allosteric inhibitor of SHP2 (Src homology region 2 domain-containing phosphatase 2). By targeting both upstream (SHP2) and downstream (ERK1/2) components of the MAPK pathway, the combination aims to achieve synergistic blockade and prevent the emergence of drug resistance.
