Overview

Growth hormone-releasing peptide 6 (GHRP-6) is a synthetic hexapeptide and met-enkephalin analogue developed as a potent stimulator of endogenous growth hormone (GH) release. It acts as a growth hormone secretagogue by binding to the ghrelin receptor (formerly known as the growth hormone secretagogue receptor, GHSR), mimicking the action of ghrelin, and triggering pulsatile GH secretion from pituitary somatotrophs[1][2][5]. Unlike natural GH-releasing hormones, GHRP-6 has no sequence similarity to them and works through distinct signaling pathways involving protein kinase C activation and intracellular calcium mobilization, independent of cAMP[2]. Beyond its primary use in research for GH deficiency, it has shown potential cardioprotective effects in preclinical models—reducing doxorubicin-induced cardiac toxicity—and cytoprotective effects against multi-organ injury[4][6]. It is also explored for muscle recovery, fat metabolism enhancement, tissue repair, and possible neuroprotective benefits. Its use is prohibited in sports by anti-doping agencies due to performance-enhancing properties[1].
