Overview

**GI-108** is a fourth-generation, first-in-class bispecific fusion protein developed as a metabolic immune anticancer drug primarily for solid tumors. It fuses a **CD73-targeting antibody** with a modified **interleukin-2 variant (IL-2v/IL-2v3)**. The CD73 antibody blocks the adenosine-mediated immune evasion in tumors by inhibiting adenosine (produced via CD73/ecto-5'-nucleotidase), while the engineered IL-2 variant selectively activates **CD8+ T cells** and NK cells with minimal stimulation of regulatory T cells. This design aims to reinvigorate anti-tumor immunity even in "cold" tumors, which are resistant to existing immunotherapies and have high CD73 expression[1][2][4][5][7][8][10]. Preclinical and early clinical data indicate potent antitumor activity and low toxicity, including superior CD8+ T cell activation versus existing drugs, and confirmed efficacy in models such as triple-negative breast cancer[1]. GI-108 is being developed and clinically studied as monotherapy for advanced and metastatic solid tumors, including pancreatic cancer, non-small cell lung cancer, renal cell carcinoma, and potentially others[8][10]. It possesses synergy potential when combined with checkpoint inhibitors[2].
