GI-128 is a bispecific macrophage-engaging antibody developed by GI Innovation. It is composed of an anti-PD-L1 single domain antibody (sdAb) and an anti-LILRBs sdAb, specifically targeting LILRB1, LILRB2, and LILRB4. By simultaneously binding to PD-L1 on tumor cells and LILRB receptors on myeloid cells, GI-128 aims to reprogram immunosuppressive M2 tumor-associated macrophages into immunostimulatory M1 phenotypes and enhance macrophage-mediated phagocytosis of tumor cells. This approach is intended to overcome resistance to traditional immune checkpoint inhibitors in PD-L1-positive tumors.
