Overview

GI-270384X is an oral small molecule drug developed by GSK as a novel **inhibitor of ICAM-1 (Intercellular adhesion molecule-1) and E-selectin expression**[1][3][7]. By inhibiting these cell adhesion molecules, GI-270384X reduces leukocyte adhesion and recruitment to inflamed tissues. It demonstrated efficacy in preclinical models for attenuating experimental colitis and visceral hyperalgesia (increased pain sensitivity related to the gastrointestinal tract), particularly in models of acute inflammatory hyperalgesia and moderate colitis. However, it was less effective in models of severe or chronic colitis[5][7]. The drug was assessed in a terminated (Phase 1) clinical trial for mild to moderate ulcerative colitis, where it aimed to provide efficacy, pharmacokinetic, and safety data[2].
