Overview

GI-4000 is an investigational therapeutic cancer vaccine designed to stimulate the immune system to target and destroy cancer cells harboring mutated forms of the Ras oncogene, which are commonly found in solid tumors such as pancreatic, colorectal, and certain breast cancers. The vaccine consists of heat-killed recombinant *Saccharomyces cerevisiae* yeast engineered to express specific mutant Ras proteins (including KRAS, HRAS, and NRAS). Upon administration by subcutaneous injection, antigen-presenting cells process the yeast-based immunogen and present Ras-derived peptides via MHC class I and II pathways. This leads to activation of both cytotoxic CD8+ T-cells and helper CD4+ T-cells against tumor cells expressing mutant Ras proteins. The approach aims for a mutation-specific immune response with minimal toxicity to normal tissues. Clinical trials have shown that GI-4000 can induce antigen-specific cellular immunity in patients with advanced cancers who have failed prior therapies[1][2][5][6][7].
