GIF-1100 is a recombinant form of human Growth Inhibitory Factor (GIF), a 68-amino acid protein also classified as Metallothionein-III (MT-III). GIF is a brain-specific member of the metallothionein family that was initially identified as being significantly reduced in the brains of patients with Alzheimer's disease. Unlike other metallothioneins, GIF/MT-III has the unique ability to inhibit the growth of neonatal rat cortical neurons in vitro. GIF-1100 was developed as a potential therapeutic agent to address neurodegenerative conditions and brain injury by modulating neuronal growth and providing neuroprotection. Its mechanism of action involves the sequestration of neurotoxic metal ions, such as zinc and copper, which are implicated in oxidative stress and the formation of amyloid plaques, as well as the direct inhibition of neurotrophic factors that may cause pathological neuronal sprouting. Development was primarily led by Asahi Kasei Pharma, but the candidate did not progress to late-stage clinical trials and is currently considered discontinued.
