GIFT-21 DC is an adoptive dendritic cell immunotherapy produced by differentiating monocytes with GIFT-21, a fusion protein comprising granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-21 (IL-21). This process yields a novel dendritic cell population with a unique phenotype that combines features of both conventional and plasmacytoid dendritic cells. These cells are highly effective at cross-presenting antigens and secreting pro-inflammatory factors such as CCL2, IL-6, TNF-α, and IFN-α. In preclinical studies, GIFT-21 DCs have shown the ability to migrate to tumors, sample antigens, and prime CD8+ T cells, leading to significant tumor growth inhibition in melanoma and breast cancer models, particularly when combined with 5-fluorouracil chemotherapy.
