GIGA-564 is a fully human monoclonal antibody developed for oncology indications, specifically targeting metastatic or locally advanced solid tumors. Unlike first-generation anti-CTLA-4 therapies that strongly block CTLA-4’s interaction with its ligands (B7), GIGA-564 is designed to minimally block CTLA-4 and instead selectively deplete intratumoral regulatory T cells (Tregs) within the tumor microenvironment. This mechanism aims to enhance anti-tumor immune responses while reducing immune-related toxicities commonly associated with strong checkpoint inhibition. Preclinical studies have shown that GIGA-564 induces less proliferation of Tregs and causes lower toxicity compared to ipilimumab, a commercially available anti-CTLA-4 drug. The drug is administered intravenously and is currently being evaluated in Phase 1 clinical trials for patients with advanced or metastatic solid tumors who are ineligible for standard-of-care treatments or have relapsed after prior therapies.
