Overview

**Gimistotug (BGB-A445)** is a humanized IgG1κ monoclonal antibody that acts as an agonist of OX40 (TNFRSF4, CD134), a co-stimulatory receptor on T lymphocytes. Developed by BeiGene (initially by BeOne Medicines Ltd.), it selectively binds OX40 to promote proliferation and survival of memory and effector T cells, enhancing T-cell-mediated antitumor immune responses and tumor cell lysis. It is under investigation in Phase 3 trials for advanced solid tumors, including non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), and nasopharyngeal carcinoma (NPC), often in combination with tislelizumab (anti-PD-1) or chemotherapy like docetaxel, administered intravenously.[1][3][5][7]
