Ginisortamab (UCB6114) is a first-in-class, fully human monoclonal antibody developed for the treatment of advanced solid tumors, including metastatic pancreatic ductal adenocarcinoma. Its mechanism of action involves binding to and inhibiting gremlin-1, an extracellular antagonist of bone morphogenetic proteins (BMP)-2, -4, and -7. By blocking gremlin-1, ginisortamab restores BMP signaling pathways that are typically suppressed in various cancers due to high gremlin-1 expression. This restoration is believed to suppress tumor growth and potentially enhance sensitivity to chemotherapy. Ginisortamab is administered intravenously and has been evaluated as both monotherapy and in combination with standard chemotherapies such as nab-paclitaxel and gemcitabine in Phase 1/2 clinical trials for advanced solid tumors[1][2][4][5][6][7].
