Overview

Ginolimod is an orally administered small molecule sphingosine-1-phosphate (S1P) receptor modulator marketed by Innovis Pharma. It is a generic formulation of fingolimod, indicated for the treatment of relapsing-remitting multiple sclerosis (RRMS). Fingolimod is a prodrug that is phosphorylated in vivo to fingolimod-phosphate, which binds with high affinity to S1P receptors 1, 3, 4, and 5. This binding induces receptor internalization and degradation, effectively acting as a functional antagonist of S1P1 receptors on lymphocytes. By blocking the S1P-dependent egress of lymphocytes from lymph nodes, Ginolimod reduces the number of circulating lymphocytes available to infiltrate the central nervous system, thereby mitigating the inflammatory processes that lead to nerve damage and clinical relapses in patients with multiple sclerosis.
