Ginsenoside Rg5 is a rare tetracyclic triterpenoid saponin primarily derived from red ginseng (*Panax ginseng*) through the steaming and drying process of white ginseng. It has demonstrated significant pharmacological potential, particularly as an anti-cancer agent. Mechanistically, Rg5 has been shown to inhibit colorectal cancer progression by acting as an autophagy inhibitor; it specifically targets and inhibits the ATP6V1A subunit of the vacuolar-type H+-ATPase (V-ATPase), which blocks lysosomal degradation and autophagic flux, ultimately triggering apoptosis. Beyond oncology, Rg5 is researched for its anti-inflammatory, neuroprotective, and anti-diabetic effects, often modulating pathways such as NF-κB and PI3K/Akt.
