GIPC1-LNPs are a therapeutic delivery system utilizing lipid nanoparticles (LNPs) to restore the expression of GIPC1 (PDZ Domain Containing Family Member 1), a tumor suppressor frequently downregulated in colorectal cancer (CRC). By increasing GIPC1 levels, the treatment inhibits CRC cell proliferation, migration, and invasion. The mechanism involves GIPC1-mediated downregulation of the E3 ubiquitin ligase TRIM21, which prevents the ubiquitination and degradation of TTC7B (tetratricopeptide repeat domain 7B). Stabilized TTC7B subsequently inhibits the mTOR and NF-κB signaling pathways, which are critical for tumor progression and chemoresistance. Preclinical evidence suggests that GIPC1-LNPs can enhance the efficacy of standard chemotherapies such as 5-fluorouracil, oxaliplatin, and irinotecan.
