Overview

GIPR-Ab + GLP-1 is an experimental combination therapy consisting of a monoclonal antibody antagonist targeting the glucose-dependent insulinotropic polypeptide receptor (GIPR) and a glucagon-like peptide-1 (GLP-1) receptor agonist. The GIPR antibody blocks the action of endogenous GIP at its receptor, while the GLP-1 agonist activates the GLP-1 receptor. Preclinical studies in obese mice and nonhuman primates have shown that this combination leads to enhanced weight loss compared to either agent alone, with improvements in body weight, fat mass, fasting serum insulin, and triglycerides. The rationale for combining these agents is based on their complementary roles in metabolic regulation—GLP-1 agonists promote satiety and enhance insulin secretion in a glucose-dependent manner, while antagonism of GIP signaling may further potentiate anti-obesity effects[1][3]. This approach is under investigation primarily for obesity treatment.
