Overview

GIPR mAb1 is a mouse anti-murine monoclonal antibody developed by Amgen as a research tool to study the glucose-dependent insulinotropic polypeptide receptor (GIPR). Unlike therapeutic candidates, mAb1 is a non-neutralizing binder that attaches to the GIPR extracellular domain with high affinity (KD of 144 pM) without competing with the natural GIP ligand or inhibiting receptor signaling. In preclinical models of diet-induced obesity, mAb1 demonstrated no therapeutic effect on body weight or metabolic parameters, serving as a negative control for the antagonistic antibody mAb2. Its characterization, including a 2.1 Å co-crystal structure, helped elucidate the molecular mechanisms of GIPR binding and informed the development of subsequent GIPR-targeted therapies for metabolic disorders.
