This is a combination therapy consisting of giredestrant (a selective estrogen receptor antagonist and degrader) plus pertuzumab and trastuzumab (HER2-targeted monoclonal antibodies). The combination is designed to simultaneously target both the estrogen receptor (ER) and HER2 pathways in breast cancer. Giredestrant blocks the cancer cell's ability to use estrogen for growth, while pertuzumab and trastuzumab attach to HER2 on cancer cells to prevent HER2-driven growth. This approach aims to overcome potential resistance mechanisms in HER2-positive, ER-positive breast cancer by addressing the bi-directional crosstalk between these pathways.
