Overview

Girentuximab is a chimeric IgG1 monoclonal antibody that targets carbonic anhydrase IX (CAIX), a cell surface antigen highly expressed in the majority of clear cell renal cell carcinomas (ccRCC) and other hypoxic tumor microenvironments. By binding to CAIX, girentuximab facilitates immune-mediated destruction of tumor cells through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). It was originally developed by Wilex AG and later licensed by Telix Pharmaceuticals for use as both an unconjugated antibody and as a radioimmunoconjugate. Girentuximab has been investigated primarily for the treatment of renal cell carcinoma, including adjuvant therapy after nephrectomy and in metastatic settings, but did not demonstrate significant improvement in disease-free or overall survival in phase 3 trials as monotherapy. The drug is also being explored in combination with radioisotopes for precision oncology applications[1][2][5][8].
