GITRL-Fc (OMP-336B11) is an investigational recombinant fusion protein developed by OncoMed Pharmaceuticals for the treatment of advanced or metastatic solid tumors. The molecule is composed of a trimeric human GITR ligand (GITRL) fused to an immunoglobulin Fc domain, designed to act as an agonist of the glucocorticoid-induced tumor necrosis factor receptor (GITR). GITR is a co-stimulatory receptor found on various T cell subsets; its activation by GITRL-Fc promotes the expansion and activity of effector T cells (CD4+ and CD8+) while potentially inhibiting the immunosuppressive function of regulatory T cells (Tregs). This dual action is intended to enhance the body's anti-tumor immune response. Clinical development reached a Phase 1a dose-escalation study, but the program was subsequently terminated following strategic shifts at OncoMed and the termination of their collaboration with Celgene.
