Overview

Givopegsomatropin is a long-acting, PEGylated recombinant human growth hormone (rhGH) developed by GeneScience Pharmaceuticals (GenSci). It consists of a 191-amino acid somatropin molecule covalently conjugated to a 20 kDa monomethoxy-polyethylene glycol (mPEG) chain. This PEGylation increases the hydrodynamic radius of the protein and reduces its renal clearance, significantly extending the half-life compared to daily rhGH injections and allowing for once-weekly subcutaneous administration. Givopegsomatropin acts as an agonist at the growth hormone receptor (GHR), triggering the JAK2/STAT5 signaling pathway to stimulate the production of insulin-like growth factor 1 (IGF-1). It was the first long-acting growth hormone approved in China (marketed as Jintrolong) for the treatment of growth failure in children with endogenous growth hormone deficiency (GHD). It is also approved for and used in the treatment of short stature associated with Turner Syndrome.
