GJB2 shRNA is a gene-silencing therapeutic candidate designed to knock down the expression of the gap junction protein beta 2 (GJB2), also known as Connexin 26. Identified through pan-cancer analysis of ion channel genes, GJB2 has been prioritized as a potential oncogene in glioblastoma (GBM). High expression of GJB2 is associated with poor prognosis and is predominantly found in malignant cell types. The shRNA-mediated knockdown of GJB2 has been shown to impair cell viability, spheroid formation, and the length of tunneling nanotubes in patient-derived GBM cell lines. In vivo studies using mouse xenograft models demonstrated that GJB2 shRNA treatment significantly prolonged survival, suggesting its potential as a novel therapeutic approach for GBM by disrupting tumor cell connectivity.
