Overview

GK NAM is a small molecule negative allosteric modulator of glucokinase (GK) currently in early-stage development by Hua Medicine. Glucokinase acts as a critical glucose sensor in the liver and pancreatic beta cells, regulating insulin secretion and glucose metabolism. While glucokinase activators (GKAs) like dorzagliatin are used to treat type 2 diabetes, negative allosteric modulators (NAMs) are designed to reduce the enzyme's activity or sensitivity. This mechanism is primarily investigated for the treatment of metabolic conditions characterized by hyperinsulinemia and hypoglycemia, such as congenital hyperinsulinism (CHI) or persistent hyperinsulinemic hypoglycemia of infancy (PHHI), where overactive glucokinase leads to excessive insulin secretion.
