GKIND009A is a small molecule potassium-competitive acid blocker (P-CAB) being developed by China Resources Guokang Pharmaceutical. It is designed to treat reflux esophagitis by inhibiting the gastric H+/K+-ATPase enzyme (the proton pump). Unlike traditional proton pump inhibitors (PPIs) that require acid activation and bind covalently, P-CABs like GKIND009A competitively inhibit the pump by binding to the potassium-binding site. This mechanism allows for a more rapid onset of action, longer-lasting acid suppression, and better control of nocturnal acid breakthrough. The drug is currently in the IND-enabling (pre-clinical) stage of development.
