GL-ONC1 + cisplatin is an investigational combination therapy consisting of **GL-ONC1** (olvimulogene nanivacirepvec), a genetically engineered, attenuated oncolytic vaccinia virus, and **cisplatin**, a platinum-based chemotherapeutic agent. GL-ONC1 selectively infects and lyses tumor cells, leading to direct oncolysis and stimulation of antitumor immune responses. The virus has been engineered to express marker genes for imaging and diagnostic monitoring. GL-ONC1 induces both local and systemic immune activation against tumor antigens, including increased infiltration of CD8+ T cells and pro-inflammatory immune modulation. Cisplatin is a well-established DNA crosslinking agent that induces apoptosis of rapidly dividing cells. The combination of GL-ONC1 with cisplatin aims to use the oncolytic and immunogenic properties of the virus with the cytotoxic effects of cisplatin, potentially providing synergistic antitumor efficacy. This combination is under investigation primarily for locoregionally advanced head and neck cancer. Early-phase studies indicate the regimen is well-tolerated with documented evidence of tumor colonization by the virus, oncolysis, and induction of antitumor immune responses[1][3][9].
