GL0469 is a preclinical small-molecule Bax (Bcl-2-associated X protein) activator being developed for the treatment of breast cancer, including estrogen receptor (ER)-negative breast cancer (ENBC) and triple-negative breast cancer (TNBC). It was discovered through the chemical optimization of progenitor Bax activators (such as GL0385 and GL0388) by researchers at the Louisiana State University Health Sciences Center and the University of Texas Medical Branch. GL0469 promotes apoptosis in cancer cells by activating Bax, which restores the anti-apoptotic/pro-apoptotic balance, leading to the release of cytochrome C, activation of caspase-3, cleavage of PARP, and subsequent reduction of cancer cell proliferation.
