Overview

GLA-LSQ is an investigational vaccine adjuvant system composed of a nanoliposomal formulation that includes two immunological adjuvants: GLA (glucopyranosyl lipid A), a synthetic monophosphoryl lipid A–like molecule and TLR4 agonist, and LSQ, a liposomal formulation of QS-21, a saponin. It is designed to enhance immune responses to co-administered antigens by stimulating both innate and adaptive immunity. GLA-LSQ has been used in clinical trials as an adjuvant for recombinant protein vaccines against diseases such as malaria (notably with rCSP and PAMVAC antigens) and tuberculosis (with ID93 antigen). Preclinical studies have shown robust T-cell immunity and reduced pathogen burden when used with these antigens. The system was developed by the Infectious Disease Research Institute (IDRI), now known as Access to Advanced Health Institute[1][2][4][5][6].
