GLA5PR GLARS-NF1 is a controlled-release (CR) oral tablet formulation of pregabalin developed for once-daily administration. It is designed to provide sustained drug release using a three-layered tablet structure with high-viscosity polymer (PEO) in the upper and lower layers to regulate the release rate of pregabalin. The formulation aims to improve patient compliance by reducing dosing frequency compared to immediate-release pregabalin. Mechanistically, it acts as a modulator of the CACNA2D1 protein (the alpha-2-delta subunit of voltage-gated calcium channels), thereby reducing neurotransmitter release and alleviating neuropathic pain symptoms. Clinical studies have demonstrated that its efficacy and safety are non-inferior to immediate-release pregabalin for peripheral neuropathic pain, including diabetic peripheral neuropathy[1][2][4][6].
