Glabridin is a prenylated isoflavan isolated from the roots of Glycyrrhiza glabra (licorice), acting as a phytoestrogen and possessing diverse biological activities including anti-inflammatory, antioxidant, antimicrobial, anti-tumor, bone protective, neuroprotective, hepatoprotective, cardiovascular protective, anti-obesity, and anti-diabetes effects. Mechanistically, it modulates several signaling pathways (e.g., NF-κB, MAPK, Wnt/β-catenin, PI3K/AKT, AMPK), inhibits tyrosinase, P-glycoprotein, and CYP2E1, and activates peroxisome proliferator-activated receptor γ (PPARγ). It is a positive allosteric modulator of GABAA receptor, exerts platelet aggregation inhibition (implicating utility in thromboembolic disorders), and acts as a skin-whitening and cosmetic agent. Clinical use is limited by poor water solubility and bioavailability; efforts to improve delivery include nanoemulsions and microneedles[1][2][4][5][6][7].
