Overview

Glasdegib is a small molecule inhibitor of the Hedgehog signaling pathway, specifically targeting and inhibiting the transmembrane protein Smoothened (SMO). By blocking SMO, glasdegib disrupts aberrant Hedgehog pathway activation, which is implicated in the proliferation and survival of certain cancer cells. Glasdegib is primarily used in combination with low-dose cytarabine for the treatment of newly diagnosed acute myeloid leukemia (AML) in adults aged 75 years or older, or those with comorbidities precluding intensive chemotherapy. Developed by Pfizer, it was approved by the FDA in November 2018 for this indication. Clinical trials have demonstrated that glasdegib plus low-dose cytarabine improves overall survival compared to cytarabine alone[1][2][4][5][6].
