Overview
Glasmacinal (EP395) is a novel, orally available, non-antibiotic macrolide developed by EpiEndo Pharmaceuticals. It belongs to a new class of therapeutics termed "Barriolides," which are structural analogues of azithromycin designed to retain anti-inflammatory and epithelial barrier-enhancing properties while lacking antibacterial activity. By strengthening the airway epithelial barrier and reducing both neutrophilic and eosinophilic inflammation, glasmacinal aims to treat chronic respiratory diseases such as chronic obstructive pulmonary disease (COPD) and asthma without the risk of promoting antimicrobial resistance. Clinical development has progressed through Phase 2a trials for COPD, demonstrating a favorable safety profile and beneficial effects on inflammatory biomarkers.
