Overview

Glavonoid is a standardized licorice flavonoid oil (LFO) derived from the roots of *Glycyrrhiza glabra* (licorice) using a proprietary extraction process. It is primarily composed of oil-soluble prenylflavonoids, with glabridin serving as the major active constituent. While Glavonoid is commercially available as a dietary supplement marketed for the reduction of visceral fat and improvement of metabolic syndrome markers, it is currently being investigated as a therapeutic agent for transthyretin amyloidosis (ATTR). Its mechanism of action in ATTR involves the kinetic stabilization of the transthyretin (TTR) tetramer. By binding to the thyroxine-binding sites of TTR, Glavonoid prevents the dissociation of the tetramer into amyloidogenic monomers, thereby inhibiting the formation of amyloid fibrils. This stabilization approach is intended to slow the progression of amyloid deposition in organs such as the heart and peripheral nerves, particularly in patients who are ineligible for standard-of-care TTR stabilizers or gene silencers.
