GLB-008 is a preclinical molecular glue degrader (MGD) developed by Hangzhou GluBio Pharmaceutical (GluBio Therapeutics) that targets Vav guanine nucleotide exchange factor 1 (VAV1). VAV1 is a critical signaling protein primarily expressed in hematopoietic cells, where it mediates T-cell and B-cell receptor signaling. By facilitating the recruitment of VAV1 to an E3 ubiquitin ligase complex, GLB-008 induces its ubiquitination and subsequent degradation via the proteasome. This targeted depletion of VAV1 is intended to suppress aberrant immune cell activation, providing a novel therapeutic strategy for the treatment of various autoimmune diseases.
