Overview
Glecaprevir is a direct-acting antiviral agent used in combination with pibrentasvir for the treatment of chronic hepatitis C virus (HCV) infection. It is a pangenotypic inhibitor of the HCV NS3/4A protease, an enzyme essential for viral replication and maturation. By inhibiting this protease, glecaprevir prevents the cleavage of the HCV polyprotein into mature proteins necessary for viral replication. The drug demonstrates activity against all major HCV genotypes (1–6) and is effective in patients who have previously failed other NS3/4A protease inhibitors. Glecaprevir was developed jointly by AbbVie and Enanta Pharmaceuticals and is marketed as part of the fixed-dose combination product Mavyret (glecaprevir/pibrentasvir), which was first approved by the FDA in 2017[2][3][5][7].
