Overview
glembatumumab vedotin + nivolumab or pembrolizumab is a combination therapy being evaluated by Celldex Therapeutics for the treatment of advanced melanoma. Glembatumumab vedotin (CDX-011) is an antibody-drug conjugate (ADC) targeting glycoprotein NMB (gpNMB), a protein overexpressed in melanoma. It consists of a human monoclonal antibody linked to the cytotoxic agent monomethyl auristatin E (MMAE), which induces cell death by inhibiting tubulin polymerization. Nivolumab and pembrolizumab are PD-1 targeted checkpoint inhibitors that enhance the immune system's ability to recognize and attack tumor cells. This combination aims to leverage the direct cytotoxic effects of the ADC with the immune-potentiating effects of PD-1 blockade, particularly in patients with checkpoint inhibitor-refractory disease.
