Gli1-siRNA is a small interfering RNA (siRNA) designed to silence the expression of the GLI family zinc finger 1 (GLI1) gene. GLI1 is a key transcription factor and terminal effector of the Hedgehog (Hh) signaling pathway, which plays a critical role in cell proliferation, differentiation, and the maintenance of cancer stem cells. In preclinical research, particularly in glioblastoma models, Gli1-siRNA has been shown to inhibit tumorsphere formation and reduce the expression of stemness markers such as CD133, ALDH1, and Nanog. By knocking down GLI1, this agent disrupts the Hh/GLI1 signaling axis, potentially overcoming resistance to chemotherapy and preventing tumor relapse. It is primarily used as a research tool to investigate the role of the Hedgehog pathway in various cancers.
