Gli2 ASO is an antisense oligonucleotide (ASO) designed to specifically target and down-regulate the expression of glioma-associated oncogene 2 (GLI2), a key transcription factor in the Sonic Hedgehog (SHH) signaling pathway. By binding to GLI2 mRNA, the ASO facilitates its degradation, leading to reduced GLI2 protein levels and the subsequent inhibition of SHH signaling, which is often aberrantly activated in various cancers. Preclinical studies have demonstrated that Gli2 ASO treatment can inhibit tumor cell proliferation, induce apoptosis, and enhance chemosensitivity to agents like paclitaxel in models of bladder cancer, castrate-resistant prostate cancer, and hepatocellular carcinoma.
