GLIO-XS15 is an experimental therapeutic peptide vaccine formulation developed by the University Hospital Tuebingen for the treatment of newly diagnosed glioblastoma multiforme (GBM), specifically targeting HLA-A2-positive patients with MGMT-methylated tumors. The vaccine consists of multiple tumor-associated peptides (TUMAPs) combined with the novel immunomodulator XS15 (Pam3Cys-GDPKHPKSF), a water-soluble Toll-like receptor 1/2 (TLR1/2) agonist designed to enhance T-cell activation and immune response. The formulation is emulsified in Montanide ISA 51 VG and includes low-dose Humulin R (U-100 regular human insulin) as an excipient to prevent the adsorption of hydrophobic peptides to the surfaces of the delivery system. It is typically administered subcutaneously in a monthly regimen as an adjunct to standard postoperative radiotherapy and temozolomide chemotherapy.
