Glioblast-13 (GB-13) is a recombinant chimeric fusion toxin developed by Targepeutics for the treatment of glioblastoma multiforme and other solid tumors. The drug consists of a mutated human interleukin-13 (IL-13) molecule fused to a truncated version of the Pseudomonas exotoxin (PE38KDEL). It specifically targets the interleukin-13 receptor alpha 2 (IL13RA2), a cell surface receptor that is highly overexpressed in various malignancies—including high-grade gliomas, pancreatic cancer, and melanoma—but has minimal expression in normal tissues. Upon binding to IL13RA2, the molecule is internalized via endocytosis; the toxin payload then catalyzes the ADP-ribosylation of elongation factor 2 (EF-2), which halts protein synthesis and leads to apoptotic cell death.
