The glioma-associated antigen (GAA) peptide-pulsed autologous dendritic cell vaccine is an investigational personalized cellular immunotherapy developed by the UCLA Jonsson Comprehensive Cancer Center in collaboration with the National Cancer Institute. This biologic therapy utilizes a patient's own dendritic cells, which are harvested via leukapheresis and matured ex vivo. These cells are then pulsed with synthetic peptides corresponding to glioma-associated antigens (GAAs) that are frequently overexpressed in malignant gliomas. The vaccine is specifically designed for patients who are HLA-A*0201 positive. Upon administration, the dendritic cells present these antigens to the host immune system to prime and activate a cytotoxic T-lymphocyte (CTL) response against the tumor cells. It has been evaluated in Phase I clinical trials (notably NCT00612001) for the treatment of malignant gliomas, including glioblastoma multiforme.
