GLIX1 is a first-in-class small molecule drug developed for the treatment of glioblastoma multiforme (GBM), the most aggressive form of brain cancer. It is being developed by Hemispherian and is currently in late-stage preclinical development. GLIX1 selectively targets the DNA damage response in cancer cells by modulating Tet methylcytosine dioxygenase 2 (TET2) enzyme activity, which leads to an accumulation of lethal double-stranded DNA breaks specifically in tumor cells while sparing healthy tissue. The drug has demonstrated efficacy in preclinical models, including total elimination or regression of GBM tumors in mice, and shows promise for treating a broad range of cancers due to its unique mechanism that exploits vulnerabilities common to many cancer types[1][6][7]. GLIX1 is orally bioavailable, penetrates the blood-brain barrier, and has shown minimal side effects on healthy cells[5].
