GLIX40 is a first-in-class small molecule activator of the Ten-eleven translocation 2 (TET2) enzyme, currently in the discovery and optimization stage of development by Hemispherian. As part of the GLIX platform, GLIX40 aims to address epigenetic dysregulation in cancer by increasing TET2 activity and restoring levels of 5-hydroxymethylcytosine (5-hmC). This restoration of 5-hmC levels is intended to induce a DNA damage response that selectively leads to the death of cancer cells while sparing healthy tissue. The drug is being explored for its potential in treating various malignancies characterized by TET2 deficiency or altered epigenetic profiles.
