Globo-H-GM2-Lewis-y-MUC1-32(aa)-sTn(c)-TF(c)-Tn(c)-KLH conjugate vaccine

The Globo-H-GM2-Lewis-y-MUC1-32(aa)-sTn(c)-TF(c)-Tn(c)-KLH conjugate vaccine is a polyvalent cancer vaccine developed by Memorial Sloan Kettering Cancer Center (MSKCC). It is designed to target seven tumor-associated carbohydrate antigens (TACAs) that are frequently overexpressed on the surface of epithelial cancer cells: Globo-H, GM2, Lewis Y, MUC1, sialyl-Tn (sTn), Thomsen-Friedenreich (TF), and Tn. Each of these antigens is individually conjugated to the immunogenic carrier protein Keyhole Limpet Hemocyanin (KLH). The vaccine is typically co-administered with the saponin-based adjuvant QS-21 to enhance the immune response, primarily inducing IgM and sometimes IgG antibodies against the targeted antigens. It has been investigated in pilot clinical trials for patients with high-risk breast cancer and ovarian, fallopian tube, or peritoneal cancers in remission to prevent recurrence by targeting micrometastatic disease.
