Globo-H-GM2-sTn-TF-Tn-KLH is a synthetic unimolecular pentavalent cancer vaccine developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of epithelial ovarian, fallopian tube, and peritoneal cancers. The vaccine consists of five tumor-associated carbohydrate antigens (TACAs)—Globo-H, GM2 (ganglioside M2), sTn (sialyl-Tn), TF (Thomsen-Friedenreich antigen), and Tn antigen—chemically synthesized on a single peptide backbone and conjugated to keyhole limpet hemocyanin (KLH) as a carrier protein. Administered with the immunological adjuvant QS-21, the vaccine is designed to induce a polyvalent antibody response against multiple targets overexpressed on the surface of ovarian cancer cells, potentially overcoming tumor heterogeneity and antigen loss.
