Glofitamab + golcadomide is a combination regimen under investigation for the treatment of relapsed or refractory B-cell non-Hodgkin lymphoma, including diffuse large B-cell lymphoma (DLBCL). Glofitamab is a bispecific monoclonal antibody that engages T cells by binding to CD3 on T cells and CD20 on B cells, thereby directing cytotoxic T cells to kill malignant B cells. Golcadomide is an oral immunomodulatory imide drug (iMiD) that binds cereblon, modulating the activity of the E3 ubiquitin ligase complex, resulting in degradation of transcription factors that regulate both immune responses and tumor cell survival. Together, the combination is being evaluated for enhanced antitumor efficacy via dual immune activation and direct tumor cell targeting, targeting both immune evasion and tumor growth[2][5].
