A combination therapy consisting of polatuzumab vedotin, glofitamab, and obinutuzumab, often referred to as the Pola-Glofit regimen. This combination is being evaluated by University College London in the Phase II PORTAL trial as a peri-CAR-T cell treatment strategy for patients with large B-cell lymphoma (LBCL). The regimen is designed to be used either as bridging therapy before CAR-T cell infusion or as a treatment for patients who have relapsed or are refractory after CAR-T therapy. Polatuzumab vedotin is an antibody-drug conjugate targeting CD79b, glofitamab is a CD20xCD3 bispecific antibody that recruits T-cells to B-cells, and obinutuzumab is a type II anti-CD20 monoclonal antibody used to enhance B-cell depletion and mitigate cytokine release syndrome.
